In my lecture, I will present our recent research on fatty acid hydratases, a relevant class of enzymes with great biotechnological potential. I will begin by providing an overview of their general characteristics, including their occurrence in prokaryotes, their function in nature, and their catalytic activity. Hence, I will discuss in deep the postulated reaction mechanism for the hydration reaction, along with its regio- and stereoselectivity. The most common substrates of fatty acid hydratases are the C18 unsaturated fatty acids possessing a cis ∆9-10 or ∆12-13 double bonds, namely oleic, linoleic and linolenic acids, which are the main components of vegetable oils. Through enzymatic hydration, these fatty acids are transformed into their corresponding 10-hydroxy and 13-hydroxy derivatives that find several industrial applications including the production of biodegradable polymers and the synthesis of natural flavours. In the latter context, I will present our process for transforming vegetable oils into high-value natural lactones through the exploitation of the fatty acid hydratase activity of probiotic bacteria belonging to the genus Lactobacillus and Bifidobacterium. Finally, I will describe the preparation and the biotechnological application of the recombinant oleate hydratase from Lactobacillus rhamnosus and linoleate hydratase from Lactobacillus acidophilus. I will show how we demonstrated that oleate hydratase is a FADH2-dependent enzyme while linoleate hydratase is a FAD-dependent enzyme. Furthermore, I will explain how we used this discovery to develop a large-scale method for the stereoselective synthesis of (R)-10-hydroxystearic acid.
